Human Serpin A11/SERPINA11 ELISA Kit

Assay Genie's sandwich ELISA, double-antibody kit is designed for the quantitative detection of human Serpin A11 (SERPINA11) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric, double-antibody sandwich format with reactivity confirmed for human specimens. The Uniprot reference for the target protein is Q86U17, and listed synonyms include antiproteinase-like 2 and serpin peptidase inhibitor clade A member 11.

The procedure uses a pre-coated microplate. Standards, samples, and blanks are loaded and incubated at 37°C for 90 minutes, followed by addition of a biotin-labelled detection antibody (37°C, 60 minutes), HRP-Streptavidin binding (37°C, 30 minutes), and TMB substrate development in the dark for 10–20 minutes. Absorbance is read at 450 nm after adding the stop solution. The kit includes a lyophilised standard, biotin-labelled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, and a plate sealer.

Reported linearity in serum (n=5) is 88–103% at 1:2, 87–103% at 1:4, and 85–104% at 1:8; EDTA plasma shows 83–96%, 84–100%, and 84–101%, and heparin plasma shows 82–95%, 81–90%, and 92–96% at the same dilutions. Average recovery is 96% in serum (range 95–102%), 95% in EDTA plasma (range 88–99%), and 93% in heparin plasma (range 97–104%).
