Description

From Assay Genie, this competitive ELISA kit enables the quantitative measurement of human Serine/threonine-protein kinase SIK2 (SIK2) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay is specific to natural and recombinant human SIK2 and provides a detection range of 0.312-20 ng/ml with a sensitivity of 0.15 ng/mL. Precision has been characterised by the manufacturer, with an intra-assay CV of 5.3% and an inter-assay CV of 8.7%. Linearity has been evaluated in serum, EDTA plasma and heparin plasma across dilution factors from 1:2 to 1:16, and recovery averages 92% (range 86-98%) in serum and 94% (range 88-100%) in plasma.

SIK2 (also known as Qin-induced kinase, Salt-inducible kinase 2, SIK-2, KIAA0781, QIK and SNF1LK2; Uniprot Q9H0K1) is a serine/threonine kinase of the CAMKL family, related to AMPK, and is localised to the cytoplasm where it interacts with and phosphorylates TORC2/CRTC2. Functionally, SIK2 phosphorylates Ser-794 of IRS1 in insulin-stimulated adipocytes, potentially modulating the efficiency of insulin signal transduction, and inhibits CREB activity by phosphorylating and repressing the CREB-specific coactivators TORC1-3. Dysregulation of SIK2 has been linked to diabetes, cancer and inflammatory diseases, making it a candidate therapeutic target and biomarker for disease progression.
