SQSTM1 functions as a selective autophagy receptor and scaffold protein, linking polyubiquitinated cargo to LC3-positive autophagosomes. It also participates in NF-κB signaling and oxidative stress responses, which is why accurate quantitation of p62 levels is used as a readout of autophagic flux in cell biology and in disease models such as Paget's disease of bone, neurodegeneration, and several cancers.
