Human Semaphorin 4D/SEMA4D ELISA Kit

A high-quality Sandwich ELISA kit from Assay Genie, designed for the accurate quantification of human Semaphorin 4D (SEMA4D) in research samples. The assay uses a double-antibody sandwich format with a colorimetric TMB readout read at 450 nm, and is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported performance characteristics from Assay Genie include a sensitivity of 75 pg/mL and a quantification range of 125-8000 pg/mL. Linearity was tested at 1:2, 1:4 and 1:8 dilutions: serum 87-100% / 85-105% / 92-100%, EDTA plasma 86-98% / 85-99% / 82-100%, and heparin plasma 82-98% / 81-90% / 85-96%. Spike-and-recovery values (n=5) are 90-102% (average 96%) in serum, 89-100% (average 97%) in EDTA plasma, and 88-104% (average 92%) in heparin plasma. Unopened kits are stored at 2-8°C for 12 months.

The protocol involves a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-streptavidin (SABC) step, and 10-20 minutes of TMB development in the dark before stop solution and absorbance reading. Semaphorin 4D is involved in immune response regulation and cell migration, and is of research interest in inflammation, autoimmune disease, and cancer.
