Human Secretogranin 2/Chromogranin C ELISA Kit

Assay Genie supplies a Sandwich ELISA designed for the accurate detection of Secretogranin 2/Chromogranin C levels in human specimens. The double-antibody Sandwich ELISA format is read colorimetrically by measuring absorbance at 450 nm after a TMB substrate reaction. The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and reactivity is limited to human samples.

Stated performance figures include a sensitivity of 0.75 ng/mL and an assay range of 1.25–80 ng/mL. Linearity values for serum at 1:2, 1:4 and 1:8 dilutions are 80–90%, 86–101% and 83–101%; for EDTA plasma 87–95%, 82–94% and 90–105%; and for heparin plasma 91–105%, 85–96% and 82–98%. Recovery averages are 96% for serum (range 86–102%), 93% for EDTA plasma (range 86–101%) and 93% for heparin plasma (range 86–100%). Unopened kit components are stored at 2-8°C for 12 months.

The protocol involves equilibration of reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, colour development with TMB in the dark for 10–20 minutes, and a stop reaction followed by immediate absorbance reading at 450 nm.
