Beyond oncology, investigators apply this Assay Genie kit to track sCD44 dynamics during immune cell activation, wound healing models, and hepatic injury studies, where elevated circulating levels signal extracellular matrix remodeling. The standard curve typically spans approximately 0.16 to 10 ng/mL, with a limit of detection in the low pg/mL range, accommodating both healthy donor sera and patient cohorts with elevated shedding.

Sample preparation guidance includes collecting whole blood in serum separator or EDTA plasma tubes, allowing 30 minutes at room temperature for clot formation, then clarifying by centrifugation at 1000 x g for 15 minutes. Aliquot cleared supernatant into single-use cryovials and store frozen at -80 °C; avoid more than two freeze-thaw cycles to preserve epitope integrity. Cell culture supernatant samples generally require 2- to 5-fold dilution in the provided buffer to fall within the calibrated range.

Each 96-well plate supports an 8-point standard curve in duplicate, dedicated blank wells, and up to 40 unknown samples run in duplicate. Intra-assay precision is typically below 8% CV and inter-assay precision below 12% CV, consistent with sandwich ELISA performance. Researchers should confirm parallelism between serially diluted samples and the recombinant standard before reporting absolute concentrations, and reserve streptavidin-HRP aliquots at -20 °C away from light to maintain enzymatic activity. Provided reagents are intended for research use only.
