Description

A sandwich ELISA, double antibody kit from Assay Genie designed for the quantitative measurement of human SCARB1 (Scavenger Receptor Class B Member 1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric readout and is intended for immunology research. SCARB1 functions as a receptor for high-density lipoprotein (HDL) and plays a role in cholesterol metabolism and lipid transport, and the protein has been implicated in cardiovascular disorders, metabolic syndromes, and atherosclerosis.

The kit has a reported sensitivity of 0.188 ng/mL and a standard curve range of 0.313–20 ng/mL. Linearity in spiked samples (n=5) was 86–105% at 1:2, 89–104% at 1:4 and 85–97% at 1:8 for serum; 89–101%, 84–95% and 82–96% for EDTA plasma; and 80–94%, 80–95% and 87–99% for heparin plasma. Spike-and-recovery was 87–100% (average 92%) for serum, 90–98% (average 94%) for EDTA plasma and 88–104% (average 98%) for heparin plasma. Unopened components should be stored at 2–8°C for 12 months.

The procedure begins with reagent equilibration, followed by a 90-minute sample incubation at 37°C on the pre-coated plate, a 60-minute biotin-labelled detection antibody incubation at 37°C, a 30-minute HRP-Streptavidin (SABC) incubation at 37°C, colour development with TMB substrate for 10–20 minutes in the dark, and absorbance reading at 450 nm after addition of stop solution. Listed synonyms for the target include SCARB1, CD36L1, CLA1, HDLQTL6 and SR-B1, and the associated UniProt entry is Q8WTV0.
