Description

A competitive ELISA assay from Assay Genie designed for the quantification of salusin alpha in human biological samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Salusin alpha is described as a novel peptide hormone implicated in regulating blood pressure, energy metabolism, and insulin sensitivity, and it has been linked to hypertension, obesity, and diabetes [S1].

The assay is a Competitive ELISA, Coated with Antigen, with a reported sensitivity of 46.875 pg/mL and a range of 78.125-5000 pg/mL [S1][C0]. Detection is colorimetric, with absorbance read at 450 nm following TMB substrate incubation [S1]. Reported linearity for serum (n=10) is 86-99% at 1:2, 82-101% at 1:4, and 82-95% at 1:8; EDTA plasma (n=10) shows 86-103%, 82-95%, and 81-99% at the same ratios, while heparin plasma (n=10) shows 85-95%, 86-99%, and 81-98% [S1]. Recovery ranges are reported as 85-99% (average 93%) for serum, 85-101% (average 95%) for EDTA plasma, and 87-105% (average 95%) for heparin plasma [S1].

The procedure involves equilibrating TMB substrate, adding 50 µL of standard or sample with 50 µL of biotin-labeled antibody per well, incubating at 37°C for 45 minutes, washing five times, developing colour in the dark at 37°C for 10-20 minutes, stopping the reaction, and reading absorbance at 450 nm [S1]. The kit is stored at 2-8°C (sealed) and should not be cryopreserved [S1].
