Innate immune signaling through the receptor for advanced glycation end products (RAGE) is mediated in part by S100A12 (also called calgranulin C or EN-RAGE), a calcium-binding protein of the S100 family that is secreted by activated neutrophils and elevated at sites of acute and chronic inflammation. In research settings, circulating S100A12 is commonly studied as a biomarker in rheumatoid arthritis, Kawasaki disease, inflammatory bowel disease, vasculitis, sepsis, atherosclerosis, and type 2 diabetes, where it serves as a readout of neutrophil degranulation and chronic innate immune activation.

The Assay Genie Human S100A12 ELISA Kit is configured as a quantitative two-antibody immunoassay supplied as a complete microplate kit, with pre-coated strips, detection antibody, streptavidin-HRP conjugate, substrate solution, wash buffer, stop solution, and a calibrator used to build the standard curve. The workflow accepts a small human sample volume per well, and after incubation with the detection reagents the absorbance signal is read and interpolated against the calibrator dilution series to determine the S100A12 concentration in each unknown.

Reconstituted reagents are stored refrigerated between uses, while unused strips are returned to the foil pouch with desiccant and kept at cool ambient temperature until the printed expiration on the kit box. The kit is intended for research use only in laboratory investigation of pro-inflammatory signaling, neutrophil biology, and disease-associated biomarker profiling, and is not validated for clinical diagnostic procedures.
