Description

Assay Genie supplies a sandwich ELISA kit built on a double-antibody format for the quantitative measurement of human RRM1 (Ribonucleoside-diphosphate reductase large subunit) in research samples. Validated sample types are serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. RRM1 is described as a key enzyme involved in DNA synthesis and repair, and dysregulated RRM1 levels have been linked to diseases including cancer and neurodegenerative disorders, making the molecule a candidate biomarker for research into disease pathogenesis and treatment development.

Linearity across dilutions (1:2, 1:4, 1:8) is reported for serum (88-101%, 94-105%, 87-105%), EDTA plasma (82-92%, 83-98%, 92-99%) and heparin plasma (83-98%, 80-97%, 80-99%). Recovery ranges are given as 88-100% (average 97%) for serum, 85-99% (average 92%) for EDTA plasma and 89-105% (average 98%) for heparin plasma.

The assay workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labelled detection antibody is then added (37°C, 60 minutes), followed by HRP-Streptavidin (37°C, 30 minutes), TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Unopened kit components are stored at 2-8°C for 12 months.
