Description

Assay Genie's sandwich ELISA is designed for the precise measurement of human Rho GTPase-activating protein 21 (ARHGAP21) in serum and plasma samples, and is additionally compatible with tissue homogenates, cell culture supernates and other biological fluids. The colorimetric assay detects natural and recombinant human ARHGAP21 and is supplied for research use only.

The detection range is 15.6–1000 pg/mL, with a sensitivity of 8.9 pg/mL. Intra-assay CV is 4.5% and inter-assay CV is 7.9%. Across dilution factors of 1:2, 1:4, 1:8 and 1:16, linearity in serum ranges from 89–119%, in EDTA plasma from 109–127%, and in heparin plasma from 95–116%. Recovery is 94% (88–100%) in serum and 96% (90–102%) in plasma.

ARHGAP21 functions as a GTPase-activating protein (GAP) for RHOA and CDC42, acts as a downstream partner of ARF1, and is required for CTNNA1 recruitment to adherens junctions. The protein interacts with GTP-bound ARF1 and ARF6 and with CTNNA1, and localises to the Golgi apparatus membrane, cell junctions, cytoplasmic vesicle membranes, the cytoplasm and the cytoskeleton. Observed molecular weight is 89 kDa and predicted molecular weight is 218 kDa (UniProt Q5T5U3).
