Description

Assay Genie offers a Sandwich ELISA Kit designed for the detection of human Retinoblastoma-like protein 1 (RBL1). The kit is reactive with human samples and is suited to research applications, including investigations into cancer biology and tumour suppression. RBL1 is similar in sequence and function to the retinoblastoma 1 (RB1) tumour suppressor protein, and its dysregulation has been linked to various cancers, making it a valuable biomarker for studying cancer development and progression.

The kit employs a sandwich ELISA format with reactivity for human analytes. It is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, and shows specificity for natural and recombinant human Retinoblastoma-like protein 1. Performance characteristics include a detection range of 1.56-100ng/mL, a sensitivity of 0.98ng/mL, an intra-assay CV of 6.2% and an inter-assay CV of 11.4%. Linearity testing across dilutions from 1:2 to 1:16 yielded ranges of 92-104% for serum, 103-112% for EDTA plasma and 107-118% for heparin plasma. Spike-and-recovery testing returned an average of 116% (range 110-120%) for serum and 118% (range 112-120%) for plasma.

RBL1, also known as 107 kDa retinoblastoma-associated protein, pRb1 and p107, is a nuclear protein catalogued under UniProt accession P28749. It functions as a key regulator of cell cycle entry and is directly involved in heterochromatin formation, recruiting histone methyltransferases such as KMT5B and KMT5C to maintain overall chromatin structure and mediate epigenetic transcriptional repression. RBL1 controls histone H4 'Lys-20' trimethylation and is a potent inhibitor of E2F-mediated trans-activation. It may act as a tumour suppressor. For research use only.
