Description

A colorimetric, double-antibody sandwich ELISA from Assay Genie, developed for the quantitative detection of human reticulocalbin-1 in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reticulocalbin-1 is a calcium-binding protein linked to intracellular calcium homeostasis, protein folding, secretion, and cell signalling, with dysregulation reported in cancer, neurodegenerative disorders, and diabetes, making it a relevant target for biomarker and therapeutic research.

Performance characteristics include a sensitivity of 0.375 ng/mL and an assay range of 0.625-40 ng/mL. Recovery in serum is 93-105% (average 100%), in EDTA plasma 85-102% (average 94%), and in heparin plasma 91-101% (average 96%). The kit is intended for storage at 2-8°C for 12 months.

The assay follows a six-step protocol: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and stop reaction followed by absorbance reading at 450 nm. Sample preparation guidance covers serum (clot, then centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant collection, centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenate, and cell lysate workflows.
