Description

Assay Genie's Sandwich ELISA kit is designed for the quantitative detection of RET (Proto-oncogene tyrosine-protein kinase receptor Ret) in human samples. The kit uses a colorimetric, double-antibody sandwich format and is intended for research applications in cell biology, including studies of RET's role in cell signalling pathways and its links to nervous system development and to various cancers.

Key performance specifications include a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Spiked recovery averages 97% in serum, 94% in EDTA plasma, and 97% in heparin plasma. Linearity has been confirmed across dilutions of 1:2, 1:4, and 1:8 in serum, EDTA plasma, and heparin plasma. Reconstituted components are stored at 2-8°C for up to 12 months.

The assay follows a six-step protocol: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant, and cell lysates, with technical support available for additional sample types.
