Description

From Assay Genie, this Sandwich ELISA Kit enables the quantitative measurement of Regulatory-associated protein of mTOR in human samples. Using a double-antibody sandwich ELISA detection method, the assay is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported performance values include a sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL. Components are intended to be stored at 2-8°C for 12 months. The target analyte is also known as RPTOR or Raptor and corresponds to Uniprot entry Q8N122.

The assay is run in six steps: equilibrate reagents and TMB substrate, then load standards, samples and a zero control onto the pre-coated plate and incubate at 37°C for 90 minutes; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; add TMB substrate and incubate in the dark for 10-20 minutes; add stop solution and read absorbance at 450 nm. Reported linearity in serum at 1:2, 1:4 and 1:8 dilutions is 89-101%, 83-97% and 83-89%; in EDTA plasma, 85-105%, 83-100% and 80-99%; and in heparin plasma, 87-101%, 88-99% and 82-89%. Reported recovery averages are 96% for serum (range 86-104%), 91% for EDTA plasma (range 86-96%) and 96% for heparin plasma (range 88-101%).
