Description

A sandwich ELISA kit from Assay Genie for the quantitative measurement of Human Regenerating Islet-Derived Protein 3 Alpha (REG3A). The assay is reactive with human samples and is designed for use with serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples, with recommended storage at 2-8°C for 12 months.

Reported performance characteristics include a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Linearity in serum, EDTA plasma and heparin plasma spans roughly 82-104% across 1:2, 1:4 and 1:8 dilutions. Spike-and-recovery performance is reported as 85-105% for serum, 92-105% for EDTA plasma, and 89-103% for heparin plasma, with averages of 94%, 97% and 96% respectively.

The procedure follows a standard sandwich ELISA workflow: equilibrate reagents and the TMB substrate, load standards and samples onto the pre-coated plate and incubate at 37°C for 90 minutes, incubate with a biotin-labeled detection antibody at 37°C for 60 minutes, incubate with HRP-Streptavidin (SABC) at 37°C for 30 minutes, develop with TMB substrate in the dark for 10-20 minutes, add stop solution and measure absorbance at 450 nm. REG3A is described in the source literature as a protein involved in tissue regeneration, immune regulation, wound healing, inflammatory responses and tissue repair, and is cited as a biomarker of interest in inflammatory bowel disease, liver disease and cancer research.
