Human RECK ELISA Kit

A sandwich ELISA, double antibody assay from Assay Genie designed for the quantitative measurement of human RECK (Reversion-inducing-cysteine-rich protein with kazal motifs) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method and is suited to research applications in metabolism, including studies of tumour cell invasion and metastasis, where RECK is known to regulate matrix metalloproteinases and has been linked to various cancers.

Sensitivity is 0.375 ng/mL with a working range of 0.625–40 ng/mL. Reagents should be stored at 2–8°C for 12 months. The associated UniProt entry is O95980, and reported synonyms include ST15 and hRECK.

The assay procedure follows a six-step protocol: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody binding at 37°C for 60 minutes; HRP-streptavidin (SABC) binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and stop reaction with absorbance read at 450 nm. Reported linearity (n = 5) at 1:2, 1:4 and 1:8 dilutions is 82–95%, 82–100% and 81–88% for serum; 86–103%, 85–101% and 82–99% for EDTA plasma; and 86–105%, 85–102% and 87–103% for heparin plasma. Average recovery is 94% in serum, 95% in EDTA plasma and 95% in heparin plasma.
