Retinol Dehydrogenase 12 (RDH12) is a member of the short-chain dehydrogenase/reductase superfamily that catalyzes NADPH-dependent reduction of all-trans-retinal to all-trans-retinol within the retinal pigment epithelium. Loss-of-function mutations in the RDH12 gene are a recognized cause of Leber congenital amaurosis type 13 (LCA13) and autosomal recessive early-onset severe retinal dystrophy, making quantitative measurement of this enzyme valuable for ophthalmology and inherited retinal disease research. This Assay Genie kit supplies a precoated microplate, capture antibody, biotinylated detection reagent, streptavidin-HRP conjugate, calibrator, and substrate solution sufficient for the indicated number of determinations. Reconstituted standards and conjugate should be aliquoted and stored frozen to limit freeze-thaw cycles, while the unopened kit is held under refrigerated storage until first use. Researchers studying photoreceptor biology, retinoic acid signaling, and patient-derived retinal cell models can apply the assay in vitro to lysates prepared from cultured cells, with absorbance proportional to RDH12 levels across the supplied standard curve. The kit is intended for research use only.
