The RBBP5 protein functions as a core scaffold component of MLL/COMPASS histone methyltransferase complexes, where it is required for proper trimethylation of histone H3 at lysine 4. This chromatin modification localizes to active promoters and has been implicated in hematopoietic lineage commitment, with RBBP5 depletion impairing both myeloid and lymphoid differentiation. Elevated or dysregulated RBBP5 expression has been reported in acute myeloid leukemia and several epithelial cancers, positioning the protein as a target of interest for cancer epigenetics and biomarker discovery programs.
