The Human RARS Sandwich ELISA Kit from Assay Genie supports quantitative work in aminoacyl-tRNA synthetase biology and peripheral neuropathy research. Dominant and biallelic mutations in RARS1 have been mapped to Charcot-Marie-Tooth disease type 2N (CMT2N), an axonal neuropathy, so reproducible measurement of cytoplasmic RARS protein in serum, plasma, or cell lysate is relevant for translational biomarker and genotype-phenotype studies. The 96-well microplate is pre-coated with capture antibody; samples and recombinant standards are loaded at roughly 100 µL per well, followed by biotinylated detection antibody and streptavidin-HRP, with a combined incubation of about 3-4 hours at room temperature. An eight-point standard curve, typically covering approximately 0.3-10 ng/mL, allows interpolation of unknowns, and reported intra-assay precision is generally under 10% CV with inter-assay CV under 12%. Aliquot serum or plasma samples ahead of use and limit freeze-thaw cycles to preserve analyte integrity; allow all reagents to equilibrate before beginning the assay. Stop solution (2 N sulfuric acid) is added prior to reading absorbance at 450 nm with a 570 nm reference correction optional. For research use only, not for diagnostic procedures.
