Description

A quantitative immunoassay from Assay Genie for measuring Recombination Activating Gene 1 (RAG1) in human samples. RAG1 is a key enzyme essential for V(D)J recombination, the process that generates diversity in antigen receptors during immune system development. Dysregulation of RAG1 has been linked to various immunodeficiency disorders and autoimmune diseases, making it a valuable biomarker for research into immune system function and disease pathogenesis.

The kit uses a Sandwich ELISA, double-antibody format with colorimetric detection and is reactive with human samples, accepting serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 9.375 pg/mL with an assay range of 15.625-1000 pg/mL. Reagents are stored at 2-8°C for 12 months. Associated identifiers include UniProt P15918, KEGG K10628, and Gene ID 5896. Synonyms for the target include RAG-1, Recombination Activating Gene 1, RAG1, RNF74, RING finger protein 74, V, and DJ recombination-activating protein 1. The kit is positioned within the Epigenetics and Nuclear Signaling research area.

The assay is performed on a pre-coated microplate. Standards and samples are incubated at 37°C for 90 minutes, followed by a biotin-labelled detection antibody (60 minutes) and an HRP-Streptavidin conjugate (30 minutes). TMB substrate is added for 10-20 minutes in the dark before stop solution is applied and absorbance is read at 450 nm. Reported linearity spans 80-105% across serum, EDTA plasma, and heparin plasma at dilutions of 1:2 to 1:8. Recovery in the same matrices averages 95-99%.
