Description

A high-quality Sandwich ELISA from Assay Genie designed for the accurate detection of RAB10 levels in human samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. RAB10 is a small GTPase protein that plays a critical role in intracellular vesicle trafficking and cellular signaling pathways. Dysregulation of RAB10 has been implicated in various diseases, including cancer, neurodegenerative disorders, and metabolic disorders, making it a valuable biomarker for research into disease pathogenesis and progression.

The assay uses a Sandwich ELISA, Double Antibody detection format with a sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL. The kit is stored at 2–8°C for 12 months. Reported linearity across 1:2, 1:4, and 1:8 dilutions includes serum 89–104% / 89–102% / 88–103%, EDTA plasma 82–99% / 84–99% / 83–90%, and heparin plasma 80–96% / 84–100% / 80–98%. Recovery ranges are 92–100% for serum (average 97%), 86–102% for EDTA plasma (average 92%), and 86–105% for heparin plasma (average 100%).

The procedure begins by equilibrating reagents and TMB substrate to room temperature, then loading standards, samples, and a zero control onto the pre-coated plate for incubation at 37°C for 90 minutes. A biotin-labeled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) at 37°C for 30 minutes. TMB substrate is then added and incubated in the dark for 10–20 minutes. The reaction is stopped and absorbance is measured at 450 nm immediately.
