Pyruvate kinase (PK) catalyzes the final committed step of glycolysis, transferring a phosphate group from phosphoenolpyruvate to ADP to generate pyruvate and ATP. The liver-type and red blood cell-type isoforms are both encoded by the PKLR gene and produced by alternative mRNA splicing, so a single quantitative measurement captures both tissue-specific pools. Quantifying PKLR protein is useful in studies of hepatic carbohydrate metabolism, erythrocyte ATP balance, and the laboratory workup of hereditary non-spherocytic hemolytic anemia.

Assay Genie's microplate immunoassay supplies pre-coated wells, biotinylated detection antibody, streptavidin-HRP conjugate, recombinant PKLR standards, wash buffer, and TMB substrate in a single kit calibrated for measurement of PKLR in human tissue homogenates, cell lysates, and related sample matrices. The absorbance-based endpoint is read on a standard microplate spectrophotometer, and quantitative PKLR concentrations are interpolated from a standard curve generated on the same plate. Each kit provides enough reagents for 96 individual determinations in one plate run.

The kit is intended for research use only and should be refrigerated at 4 °C on arrival. Reconstituted standards, diluted detection antibody, and working wash buffer are kept refrigerated between plate loads to preserve reagent activity; repeated freeze-thaw cycles of the standard should be avoided by aliquoting the stock upon first thaw.
