Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of Pyruvate Dehydrogenase E1 (PDH E1) in human samples. The kit uses a double-antibody sandwich format with colorimetric detection and supports serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Performance specifications include a sensitivity of 18.75 pg/mL and an assay range of 31.25–2000 pg/mL. Reported linearity across dilutions (n = 5) is 87–99% at 1:2, 86–101% at 1:4, and 81–94% at 1:8 for serum; 86–98%, 85–99%, and 83–90% for EDTA plasma; and 86–98%, 86–100%, and 81–97% for heparin plasma. Spike recovery (n = 5) ranges from 87–103% (average 96%) in serum, 89–97% (average 93%) in EDTA plasma, and 86–102% (average 95%) in heparin plasma. The kit is stored at 2–8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after stop solution addition. PDH E1 is a key enzyme involved in the conversion of pyruvate to acetyl-CoA within mitochondria, playing a crucial role in cellular energy production; dysregulation of PDH E1 activity has been linked to metabolic disorders, neurodegenerative diseases, and certain types of cancer.
