Pyruvate dehydrogenase alpha (PDHa) is the E1 catalytic component of the mitochondrial pyruvate dehydrogenase complex, the rate-limiting enzymatic gateway that decarboxylates pyruvate into acetyl-CoA for entry into the TCA cycle. The Assay Genie PDHa ELISA Kit delivers quantitative colorimetric measurement of this subunit in human specimens, supporting research into aerobic glucose oxidation, lactate homeostasis, mitochondrial dysfunction, and the metabolic reprogramming observed in cancer, neurodegeneration, and inborn errors of metabolism.

The kit accommodates 96 assays and supports analysis of multiple human specimen types, including tissue homogenate supernatants and serum- or plasma-derived samples collected for research purposes. Reagents are supplied in individually sealed vials and should be handled under refrigerated storage conditions, with the microplate sealed and protected from moisture between runs to preserve antibody binding activity.

Typical research applications include biomarker quantification in patient cohorts with suspected mitochondrial disease, monitoring of PDH activity in cell culture models of metabolic stress, and measurement of PDHa levels in tumor lysates from Warburg-effect studies. The assay readout is compatible with standard microplate readers, and the kit is intended for research use only. Reconstituted standard and detection antibody vials are intended for single-use aliquots to avoid repeated freeze-thaw cycles.
