Pyruvate carboxylase (PC), encoded by the PC gene on chromosome 11q13.4-q13.5 (UniProt P11498), is a biotin-dependent homotetrameric enzyme confined to the mitochondrial matrix. Each subunit carries three functional domains — biotin carboxylase, carboxyltransferase, and biotin carboxyl carrier — and the enzyme catalyzes the ATP-dependent carboxylation of pyruvate to oxaloacetate, a reaction central to gluconeogenesis, de novo lipogenesis, glutaminolysis-driven anaplerosis of the TCA cycle, and insulin secretion. Tissue expression is highest in liver, kidney, adipose, lactating mammary gland, and pancreatic islets.

Quantitative profiling of PC protein across these tissues and matrices is enabled by this Assay Genie sandwich-format ELISA. The kit is supplied as a 96-assay configuration with a pre-coated microplate, lyophilized standards, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, stop reagent, wash buffer, and diluents. Researchers studying metabolic disease, hepatic insulin resistance, type 2 diabetes mellitus, cancer cell metabolism, and mitochondrial dysfunction can use the assay to compare PC levels between experimental conditions.

For handling, unopened components should be stored refrigerated; the lyophilized standard, biotinylated antibody, streptavidin-HRP, substrate, and stop reagent are commonly held at 4 °C or frozen at −20 °C, while working aliquots should be protected from repeated freeze-thaw cycles to preserve activity. The assay is validated for research use only and is for in vitro laboratory applications.
