Assay Genie supplies this kit as a quantitative Sandwich ELISA built on a pre-coated 96-well microplate, biotinylated detection antibody, avidin-HRP conjugate, and TMB substrate that yields a yellow product read at 450 nm after the stop solution is added. PYGB is the brain-type glycogen phosphorylase isozyme (gene ID 5834), one of three human isoforms alongside the muscle (PYGM) and liver (PYGL) forms; in the central nervous system it is expressed predominantly in neurons and astrocytes, where it acts as the rate-limiting enzyme of glycogen catabolism and is allosterically activated by rising intracellular AMP. Aberrant PYGB activity has been reported in published studies on Alzheimer's disease, temporal lobe epilepsy, stress-induced depression, several cancers, and cardiovascular and metabolic disorders, supporting its use as a quantitative marker in neurology, metabolism, and oncology research workflows.

A recombinant human PYGB calibrator series is provided so the operator can build a four-parameter logistic (4-PL) standard curve, with typical detection sensitivity around 9.8 pg/mL and a working range of 15.6-1000 pg/mL on serum, plasma, tissue homogenates, and cell culture supernatant. The full workflow takes roughly 3 hours of room temperature incubation. Unopened kit components should be stored at 2-8 °C, the standard stock at -20 °C, and repeated freeze-thaw cycles of prepared reagents should be avoided. For research use only, not for diagnostic or clinical applications.
