Pyrroline-5-carboxylate reductase 1 (PYCR1) is the mitochondrial enzyme that catalyzes the NAD(P)H-dependent reduction of pyrroline-5-carboxylate to proline, the terminal step of de novo proline biosynthesis. Assay Genie's ELISA Kit provides quantitative measurement of human PYCR1, supporting research into proline metabolism, redox balance, and connective tissue disorders such as autosomal recessive cutis laxa type IIB.

Built around a pre-coated microplate, the kit supplies matched capture and detection antibodies, streptavidin-enzyme conjugate, chromogenic substrate, stop reagent, wash buffer, and a recombinant PYCR1 standard set for curve fitting. Up to 96 individual reactions can be run in parallel on a removable-strip plate, allowing flexible batch sizes without wasting unused wells. The protocol is straightforward: dilute standards and samples, incubate, wash, develop, and read absorbance to interpolate unknown concentrations against the standard curve.

Intended for research use only, the assay supports biomarker work in human cohorts. Cancer researchers studying melanoma, breast carcinoma, and other PYCR1-overexpressing tumor types can compare circulating enzyme levels against healthy controls, while dermatology groups can use the assay to track mitochondrial enzyme abundance in affected families. The kit is also useful for cell culture studies where proline auxotrophy rescue is being used as a functional readout.

Storage and handling: keep sealed plates and dry reagents refrigerated; reconstituted standards and diluted detection antibody should be frozen at -20°C for stability across multiple freeze-thaw cycles. Allow all components to reach room temperature before use, and shield the substrate vial from light. Each kit ships with a printed protocol and Certificate of Analysis documenting lot-specific validation.
