Human PTPRF/Receptor-type tyrosine-protein phosphatase F ELISA Kit

This Sandwich ELISA (double antibody) from Assay Genie is designed for the quantitative detection of human Receptor-type tyrosine-protein phosphatase F (PTPRF), also known as LAR or Leukocyte common antigen related. The assay is intended for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and employs a colorimetric detection method with a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. The kit is stable at 2–8°C for 12 months and the associated UniProt entry is P10586.

PTPRF is described in the literature as a key regulator of cell signalling pathways with roles in diverse physiological processes; its dysregulation has been linked to cancer, autoimmune disorders and neurological conditions, supporting its use as a potential biomarker for disease development and progression. Assay performance was evaluated by linearity and spike-and-recovery testing. Linearity at 1:2, 1:4 and 1:8 dilutions was 88–105%, 85–103% and 90–102% for serum (n=5); 82–98%, 83–98% and 82–99% for EDTA plasma (n=5); and 80–95%, 81–100% and 81–97% for heparin plasma (n=5). Spike-and-recovery averaged 95% for serum (range 87–105%), 94% for EDTA plasma (range 89–97%) and 96% for heparin plasma (range 89–103%).

The assay procedure involves equilibrating reagents and TMB substrate to room temperature; a primary incubation at 37°C for 90 minutes to allow antigen binding; biotin-labelled detection antibody incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes; TMB substrate colour development in the dark for 10–20 minutes; and a stop step with absorbance read at 450 nm.
