Human PTP1B ELISA Kit

Designed by Assay Genie as a double-antibody sandwich ELISA with colorimetric detection, this kit enables the quantitative measurement of human Protein Tyrosine Phosphatase 1B (PTP1B, also referred to as PTPN1; Uniprot P18031) in research samples [C0, S1, S2, S3]. The assay delivers a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL [S1, S2, S3]. PTP1B is a critical enzyme that regulates cellular signalling pathways by dephosphorylating specific tyrosine residues on target proteins, and its dysregulation has been linked to diabetes, obesity, and cancer [S1, S2, S3].

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with the kit intended for storage at 2-8°C for 12 months [S1, S2, S3]. The protocol runs through six steps: equilibrate reagents and TMB substrate to room temperature; load standards, samples and blanks on the pre-coated plate and incubate at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; bind HRP-Streptavidin (SABC) at 37°C for 30 minutes; develop colour with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm [S1, S2].

Validation data are provided for linearity and recovery across serum, EDTA plasma and heparin plasma. Reported linearity spans 91-102%, 88-100% and 84-96% in serum at 1:2, 1:4 and 1:8 dilutions (n=5), with EDTA plasma at 87-105%, 85-99% and 82-100%, and heparin plasma at 87-105%, 82-96% and 81-96% [S1, S2]. Recovery ranges were 86-105% (serum), 89-103% (EDTA plasma) and 88-104% (heparin plasma), with corresponding averages of 94%, 93% and 92% (n=5) [S1, S2, S3].
