Human PTN/Pleiotrophin ELISA Kit

A sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of pleiotrophin (PTN) levels in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. The assay uses a double-antibody sandwich format with colorimetric detection, a sensitivity of 18.75 pg/mL, and a working range of 31.25–2000 pg/mL.

Pleiotrophin is a secreted growth factor involved in cell growth, migration, differentiation, and survival, and is studied as a biomarker in contexts such as cancer, neurodevelopmental disorders, and bone regeneration. Reported linearity falls broadly between 80% and 105% across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with average spike-recovery values of 94% (serum), 95% (EDTA plasma), and 98% (heparin plasma). Unopened kit reagents are stored at 2–8°C for up to 12 months.

The workflow follows a standard sandwich protocol: equilibrate reagents and TMB substrate to room temperature, load standards, samples and blanks onto the pre-coated plate and incubate at 37°C for 90 minutes, add the biotinylated detection antibody and incubate at 37°C for 60 minutes, add HRP-streptavidin (SABC) at 37°C for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, and read absorbance at 450 nm immediately after adding the stop solution.
