Description

The PSMB9 (Proteasome subunit beta type-9) ELISA Kit from Assay Genie is a sandwich ELISA using a double-antibody format for the quantification of human PSMB9. The assay is colorimetric and reactive with human samples, with validated use in immunology research. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2–8°C for 12 months.

PSMB9 (UniProt P28065) is a key component of the proteasome complex, essential for protein degradation and maintenance of cellular homeostasis. Dysregulation of PSMB9 has been implicated in autoimmune diseases, inflammatory disorders, and cancer, underscoring its importance as a potential therapeutic target and diagnostic marker. The kit is also known by the synonyms LMP2, RING12, Proteasome chain 7, PSMB6i, and Macropain chain 7, among others.

Linearity across sample dilutions was reported as: serum (n=5) 94–105% (1:2), 82–98% (1:4), 82–96% (1:8); EDTA plasma (n=5) 89–103% (1:2), 86–100% (1:4), 80–96% (1:8); heparin plasma (n=5) 85–97% (1:2), 90–101% (1:4), 82–97% (1:8). Recovery ranges were: serum (n=5) 86–96% (average 91%), EDTA plasma (n=5) 85–99% (average 90%), heparin plasma (n=5) 85–102% (average 94%). The protocol involves primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
