PSMB3 (Proteasome 20S Subunit Beta 3) is a structural component of the constitutive 20S proteasome core particle, the catalytic chamber that degrades ubiquitin-tagged intracellular proteins in eukaryotic cells. This quantitative immunoassay from Assay Genie measures human PSMB3 protein in serum or plasma specimens using a paired-antibody capture format with a colorimetric readout at the end of the workflow. Researchers studying proteasome subunit expression, cancer biology, and protein homeostasis use the assay to track changes in circulating PSMB3 levels across patient cohorts and experimental conditions.

The kit includes a pre-coated microplate, calibrated standards, detection antibody, streptavidin-HRP conjugate, wash buffer, substrate solution, and stop reagent sufficient for 96 determinations per plate. Reconstituted standards and diluted samples are incubated in antibody-coated wells, allowing captured PSMB3 to bind the detection antibody in a sequential two-step procedure. After substrate conversion and stop solution addition, absorbance values are read on a standard microplate reader, and PSMB3 concentrations in unknown samples are interpolated from the standard curve generated on each plate.

Storage recommendations: unopened kits are kept refrigerated and protected from light until first use; reconstituted standards should be aliquoted to limit freeze-thaw cycles and stored frozen for extended use. The assay is configured for research use only and is intended for laboratory investigation of proteasome biology, biomarker discovery, and disease mechanism studies rather than clinical diagnostic applications.
