Description

Designed for the quantitative detection of PSG3 in human research samples, this double-antibody sandwich ELISA from Assay Genie is supplied as a complete kit with a colorimetric detection method. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a stated sensitivity of 0.188 ug/mL and an assay range of 0.313-20 ug/mL. All kit reagents are intended to be stored at 2-8°C, where they remain stable for 12 months.

Validation data from the manufacturer include dilution linearity across n=5 samples of each matrix. Diluted serum showed ranges of 89-105% at 1:2, 85-99% at 1:4 and 93-98% at 1:8; EDTA plasma showed 90-104%, 88-100% and 91-99% at the same dilutions; and heparin plasma showed 90-105%, 87-101% and 91-97%. Spike-and-recovery values were 86-99% (average 92%) for serum, 85-96% (average 92%) for EDTA plasma, and 90-98% (average 94%) for heparin plasma.

The workflow uses a pre-coated plate: standards and samples are incubated at 37°C for 90 minutes, followed by a 60-minute incubation with a biotinylated detection antibody, 30 minutes with HRP-streptavidin conjugate, and 10-20 minutes of TMB development in the dark, before stop solution is added and absorbance is read at 450 nm. PSG3 is a pregnancy-specific protein associated with the maternal-fetal interface and is studied as a biomarker in pregnancy-related research.
