Human PSCA/Prostate stem cell antigen ELISA Kit

A sandwich double-antibody ELISA from Assay Genie for the quantitative measurement of human Prostate Stem Cell Antigen (PSCA). The assay is reactive with human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types. According to the manufacturer's product literature, PSCA is a cell surface antigen overexpressed in prostate cancer cells, supporting its use as a biomarker for the detection and monitoring of prostate cancer in studies of the disease and related disorders.

Detection is colorimetric. The reported sensitivity is 0.188 ng/mL and the standard range is 0.313-20 ng/mL. Storage is at 2-8°C for 12 months; selected components (the dismountable ELISA microplate and the lyophilized standard) can additionally be stored at -20°C for 12 months. The kit includes a pre-coated microplate, lyophilized standard, biotin-labeled detection antibody, HRP-streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, wash buffer (25X), stop solution, plate sealer, and a technical manual.

Reported linearity in serum (n=5) is 89-103% at 1:2, 92-101% at 1:4 and 85-97% at 1:8; in EDTA plasma (n=5) 86-102%, 85-100% and 85-102%; and in heparin plasma (n=5) 88-100%, 88-98% and 94-103%. Average recoveries are 97% for serum, 92% for EDTA plasma and 96% for heparin plasma. The procedure comprises primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB substrate incubation in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution addition.
