PDI is a thiol-disulfide oxidoreductase located in the endoplasmic reticulum, where it catalyzes disulfide bond formation, isomerization, and reduction during nascent protein folding. It also operates as a molecular chaperone independent of its catalytic activity, contributing to the unfolded protein response and to redox homeostasis within the secretory pathway. Cell-surface PDI has been implicated in platelet aggregation, thrombus formation, and nitric oxide signaling, while altered PDI expression has been documented across solid tumors, viral entry mechanisms including HIV, and neurodegenerative disease models, positioning it as a candidate biomarker in cardiovascular, oncological, and neuroscience research.
