Description

Assay Genie offers a sandwich ELISA built on a double-antibody format for the quantitative measurement of protein disulfide isomerase (PDI) in human samples. The assay is configured for colorimetric readout and is calibrated across a range of 0.313–20 ng/mL, with a stated sensitivity of 0.188 ng/mL. PDI is recognised as a key enzyme in the formation and rearrangement of disulfide bonds in proteins, with dysregulation implicated in cancer, neurodegenerative disorders and cardiovascular disease, supporting its use as a biomarker for studying protein folding and related therapeutic interventions.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples. The kit is stored at 2–8 °C for 12 months. Linearity was demonstrated across serial dilutions, with serum (n = 5) returning 87–102% (1:2), 92–101% (1:4) and 81–99% (1:8); EDTA plasma (n = 5) 91–100%, 92–101% and 84–100%; and heparin plasma (n = 5) 85–105%, 83–99% and 81–96%. Recovery ranges were 85–101% for serum (average 93%), 93–104% for EDTA plasma (average 98%) and 88–105% for heparin plasma (average 95%).

The protocol proceeds through reagent and plate preparation, a primary incubation at 37 °C for 90 minutes to allow antigen binding, addition of a biotin-labelled detection antibody with incubation at 37 °C for 60 minutes, HRP-Streptavidin binding at 37 °C for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and a stop reaction with absorbance measured at 450 nm.
