Human Protein Disulfide Isomerase A3/PDIA3 ELISA Kit

From Assay Genie, this double-antibody sandwich ELISA is designed for the accurate quantification of Protein Disulfide Isomerase A3 (PDIA3) levels in human biological samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. PDIA3, also known as endoplasmic reticulum protein 57 (ERp57), is a key protein involved in the regulation of disulfide bond formation in proteins, contributing to proper protein folding and function. Dysregulation of PDIA3 has been implicated in diseases including cancer, neurodegenerative disorders, and autoimmune diseases, and it is regarded as a potential diagnostic and therapeutic target in cell biology research.

The kit uses colorimetric detection with a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. Validated linearity for spiked serum (n=5) is 92–100%, 88–105% and 89–105% at 1:2, 1:4 and 1:8 dilutions; EDTA plasma 86–99%, 82–90% and 87–96%; heparin plasma 84–100%, 87–99% and 80–104%. Mean recovery is 98% from serum (range 85–100%, n=5), 97% from EDTA plasma (range 89–100%, n=5), and 95% from heparin plasma (range 88–104%, n=5). Reagents should be stored at 2–8°C and the kit remains stable for 12 months under those conditions.

The protocol runs through reagent and plate preparation, 90-minute primary incubation at 37°C, 60-minute biotin-labelled detection antibody binding, 30-minute HRP-streptavidin (SABC) binding, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding the stop solution.
