Human Protein C Inhibitor ELISA Kit

A sandwich ELISA, double antibody assay from Assay Genie designed for the quantitative measurement of protein C inhibitor in human biological samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method with absorbance read at 450 nm, and targets protein C inhibitor (Uniprot P05154), a key regulator of blood coagulation that inhibits the activity of protein C and other factors involved in the clotting process. Imbalances in protein C inhibitor levels have been linked to thrombosis and haemorrhagic disorders, making the kit relevant to research in coagulation biology and in the developmental biology research area.

Performance characteristics include a sensitivity of 0.75 ng/mL and a working range of 1.25–80 ng/mL. Linearity has been validated across serum (85–103%, 82–100%, 86–93% at 1:2, 1:4 and 1:8 dilutions, n=5), EDTA plasma (88–100%, 86–100%, 80–96%, n=5) and heparin plasma (87–98%, 82–100%, 80–96%, n=5). Spike-and-recovery testing returned recovery ranges of 93–101% (average 97%) in serum, 89–105% (average 97%) in EDTA plasma and 94–102% (average 99%) in heparin plasma. The kit is stored at 2–8°C for 12 months.

The workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm immediately after stop solution addition. Provided reagents include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual.
