As one of seven alpha subunits forming the outer ring of the 20S proteasome core particle, Proteasome Subunit Alpha Type 1 (PSMa1) is central to ubiquitin-dependent protein degradation in human cells. Researchers studying proteasome function in multiple myeloma, Parkinson's disease, and other proteostasis-linked conditions depend on accurate quantification of this subunit.

Assay Genie's PSMa1 ELISA kit is configured for ninety-six individual determinations and measures this alpha subunit in tissue homogenates from human samples, supporting research in cell biology, oncology, and neuroscience laboratories. The protocol uses two paired antibodies to capture and detect PSMa1, producing an absorbance signal compatible with standard microplate readers found in most life science facilities. Reactivity is limited to human samples.

Reconstituted standards, detection antibody, and working solutions should be kept refrigerated between uses and discarded after the run; unused microwell strips may be returned to the foil pouch with desiccant and stored refrigerated for use within the kit's shelf life. Tissue homogenate samples should be aliquoted before storage to minimize repeated freeze-thaw cycles, which can reduce detectable PSMa1 levels in lysate preparations.

Each kit is supplied with a pre-coated microwell plate, biotinylated detection antibody, lyophilized recombinant PSMa1 standards, streptavidin-HRP conjugate, wash buffer concentrate, substrate solution, and stop solution, all packaged in single-use vials and bottles. The plate layout allows duplicate or triplicate wells for each sample, and the standard curve is run in serial dilution to support accurate interpolation across the expected range of endogenous PSMa1 concentrations.
