PRSS3, also called mesotrypsin or trypsinogen 4, is a trypsin-family serine protease notable for resisting inhibition by canonical trypsin inhibitors such as aprotinin. The protein is overexpressed in pancreatic ductal adenocarcinoma and several other solid tumours, where it has been implicated in extracellular matrix remodelling and metastatic spread. This Assay Genie two-antibody assay targets the human PRSS3 protein and supports quantitative measurement across serum-derived samples, plasma-derived samples, tissue homogenates, and conditioned media from cultured cells, addressing both clinical biomarker studies and cell-based pharmacology workflows.

Each kit contains a pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, recombinant PRSS3 reference material, and substrate buffer sufficient for 96 determinations. The workflow involves sequential incubation steps with wash cycles between reagent additions, terminating in an enzyme-catalysed colour development step whose intensity is read on a standard plate reader. A standard curve generated from serial dilutions of the provided reference material allows interpolation of sample concentrations.

Research applications include profiling PRSS3 abundance in tumour versus matched normal tissue panels, monitoring changes in protease expression during inflammatory challenge, evaluating the impact of candidate small-molecule inhibitors on cellular PRSS3 output, and stratifying biobank specimens by disease stage, as well as supporting in vitro screens of proteolytic activity. Reagents should be kept refrigerated upon receipt; the lyophilised reference standard is best reconstituted and divided into single-use aliquots stored frozen to minimise activity loss across repeated freeze-thaw cycles. The product is intended for research use only and is not validated for diagnostic procedures.
