Beyond the assay format, this Assay Genie kit targets the proPSA zymogen, the inactive single-chain precursor of kallikrein-related peptidase 3 (KLK3) that retains a short N-terminal pro-leader peptide before activation by hK2 or trypsin. In prostate cancer research, the truncated [-2]proPSA isoform accumulates preferentially in malignant peripheral zone tissue and is significantly elevated in circulation, where it contributes to the Prostate Health Index (phi) alongside total and free PSA measurements. Quantitative measurement of proPSA therefore supports investigations aimed at discriminating prostate cancer from benign prostatic hyperplasia, evaluating tumor aggressiveness in correlation with Gleason score, monitoring disease progression in cell culture and biopsy supernatant models, and screening novel diagnostic algorithms. The recommended sample volume is 100 uL per well, with the full immunoreaction sequence completed in approximately 3.5 hours at room temperature. Reconstituted standards and diluted samples should be aliquoted and stored refrigerated to limit freeze-thaw cycles, and the human-specific capture antibody shows no significant cross-reactivity with related kallikreins such as hK2 under standard conditions.
