Description

This sandwich ELISA kit from Assay Genie uses a colorimetric detection method for the quantitative measurement of human Prolyl 4-hydroxylase subunit alpha-2 (P4HA2) in serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids, with reactivity confirmed for human samples and specificity for both natural and recombinant P4HA2.

P4HA2 is an enzyme that catalyses the post-translational formation of 4-hydroxyproline in -Xaa-Pro-Gly- sequences in collagens and other proteins, playing a key role in collagen synthesis. It functions as a heterotetramer of two alpha-2 chains and two beta chains, with the alpha-2 subunit localised to the endoplasmic reticulum lumen. Dysregulation of P4HA2 has been linked to various diseases, including fibrosis, cancer, and arthritis, making it a valuable biomarker for cell biology research into collagen metabolism and related conditions.

Reported performance includes a detection range of 0.78-50 ng/mL and a sensitivity of 0.27 ng/mL. Precision values are an intra-assay CV of 4.8% and an inter-assay CV of 7.6%. Spike-and-recovery in serum averaged 95% (range 89-101%) and in plasma averaged 97% (range 91-103%), with linearity assessed across serial dilutions in serum, EDTA plasma, and heparin plasma.
