Description

Designed by Assay Genie for the in vitro quantitative determination of human Prolyl 4-hydroxylase subunit alpha-1 (P4HA1), this Sandwich ELISA kit uses a double-antibody, colorimetric detection method and is validated for use with serum, plasma, and other biological fluids. P4HA1 is a key enzyme that catalyses the post-translational formation of 4-hydroxyproline in -Xaa-Pro-Gly- sequences in collagens and other proteins, a modification essential for the proper three-dimensional folding and stability of newly synthesised procollagen chains. Dysregulation of P4HA1 has been linked to fibrosis, cancer, and wound-healing disorders, making accurate quantification of the protein valuable for studies of collagen biosynthesis and disease pathogenesis.

Performance characteristics quoted from the manufacturer include a sensitivity of 0.375 ng/ml, an assay range of 0.625-40 ng/ml, intra-assay CV <8% and inter-assay CV <10%. Spike-and-recovery in serum (n=5) was 85-100% with an average of 93%, in EDTA plasma (n=5) 86-101% with an average of 91%, and in UFH plasma (n=5) 88-102% with an average of 93%. Linearity in spiked serum spanned 86-105% across serial dilutions, with comparable results in EDTA and UFH plasma matrices. The kit is intended for research use only.
