Human Proinsulin ELISA Kit

This sandwich ELISA kit from Assay Genie is for research use only and is designed for the quantitative measurement of human proinsulin in serum, plasma, and other biological fluids. It is also referenced as the PI ELISA Kit and corresponds to Uniprot P01308. Built on a double-antibody sandwich ELISA format with a colorimetric detection method, the assay uses a pre-coated capture antibody together with a biotin-conjugated detection antibody, followed by HRP-Streptavidin binding and TMB substrate development. The resulting signal is read at 450 nm in a microplate reader.

The kit offers a measurement range of 31.25–2000 pg/ml with a sensitivity of 18.75 pg/ml. Reagents are shipped on ice packs. The assay workflow involves sequential incubations at 37°C for samples, biotin-labelled antibody, and HRP-Streptavidin conjugate, with wash steps in between, followed by TMB substrate addition and stop solution before absorbance reading. Light-sensitive reagents should be stored at 2–8°C as indicated in the kit instructions.

Sample preparation guidance covers serum, plasma (with EDTA-Na2/K2 recommended as anticoagulant), cell culture supernatant, cell lysates, and tissue homogenates, with a recommended lysis buffer of 50 mM Tris, 0.9% NaCl, 0.1% SDS at pH 7.3.
