Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of Protein C (PROC) in human samples. The kit is reactive with human specimens and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, making it a versatile tool for cardiovascular and blood coagulation research, including studies of thrombosis and sepsis.

The assay employs a colorimetric, double-antibody sandwich format with a reported sensitivity of 37.5 pg/mL and a working range of 62.5–4000 pg/mL. Linearity has been demonstrated in serum (87–101%, 88–105%, 89–103% at 1:2, 1:4 and 1:8 dilutions), EDTA plasma (85–95%, 88–100%, 82–101%) and heparin plasma (80–100%, 81–91%, 85–100%), with mean recoveries of 93% in serum, 96% in EDTA plasma and 94% in heparin plasma. Protein C is described as a vital regulator of blood clotting, and dysregulation of its levels has been implicated in various diseases, making it an important biomarker for research and clinical investigation.

Storage is at 2–8°C for 12 months. The assay procedure involves equilibrating reagents to room temperature, incubating standards and samples at 37°C for 90 minutes for antigen binding, adding a biotin-labeled detection antibody with a 37°C incubation for 60 minutes, binding HRP-Streptavidin (SABC) at 37°C for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding the stop solution.
