Description

Assay Genie provides a sandwich ELISA kit for the quantitative measurement of human Probable protein-cysteine N-palmitoyltransferase porcupine (PORCN) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay uses a colorimetric detection method and is reactive with human samples.

Reported performance characteristics include a detection range of 0.78-50ng/mL and a sensitivity of 0.23ng/mL. Intra-assay precision is given as 5.5% (CV) and inter-assay precision as 9.2% (CV). Linearity has been assessed in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16. Recovery in serum averages 91% (range 85-97%) and in plasma averages 93% (range 87-99%). The kit shows specificity for natural and recombinant human Protein-serine O-palmitoleoyltransferase porcupine.

PORCN is described as a protein-serine O-palmitoleoyltransferase that acts as a key regulator of the Wnt signaling pathway by mediating the attachment of palmitoleate to Wnt proteins, a process required for efficient binding of WNT proteins to frizzled receptors. The kit is intended for research use only and is applicable to cancer research. UniProt accession is Q9H237, with a reported molecular weight of 52kDa.
