Description
Designed for the quantitative measurement of Proadrenomedullin (Pro-ADM) in human samples, this sandwich ELISA kit from Assay Genie uses a double-antibody format with colorimetric detection. Proadrenomedullin is recognised as a key biomarker involved in regulating vascular homeostasis and inflammation, and its measurement supports research into the pathogenesis of sepsis, cardiovascular disorders, and respiratory conditions.
The assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and should be stored at 2-8°C for 12 months. Reported performance includes a sensitivity of 0.469 pmol/L and a working range of 0.781-50 pmol/L.
Validation data show serum recovery of 86-103% (average 93%), EDTA plasma recovery of 85-104% (average 94%), and heparin plasma recovery of 91-98% (average 92%). Linearity testing across 1:2, 1:4 and 1:8 dilutions gave recoveries of 87-98%, 92-101% and 92-99% respectively for serum; 86-99%, 86-97% and 86-102% for EDTA plasma; and 83-99%, 88-98% and 82-96% for heparin plasma. The procedure involves equilibrating reagents to room temperature, incubating the plate at 37°C for antigen binding (90 minutes), adding a biotin-labelled detection antibody (60 minutes), binding HRP-Streptavidin (30 minutes), developing with TMB substrate in the dark (10-20 minutes), and reading absorbance at 450 nm after adding stop solution.
