Human PRKAA1/AMPK Alpha 1 ELISA Kit

A competitive (antigen coated) enzyme-linked immunosorbent assay from Assay Genie designed for the quantitative measurement of human PRKAA1 (5'-AMP-activated protein kinase catalytic subunit alpha-1), a key regulator of cellular energy homeostasis whose dysregulation has been implicated in diabetes, cancer and metabolic disorders. The kit reacts with human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay uses microtiter wells pre-coated with PRKAA1, where analyte in the sample competes with a fixed amount of coated PRKAA1 for binding to a biotinylated detection antibody. After washing, HRP-Streptavidin (SABC) binds the captured antibody, and TMB substrate produces a colour reaction read at 450 nm; signal intensity is inversely proportional to PRKAA1 concentration. The reported detection range is 78.125-5000 pg/mL with a sensitivity of 46.875 pg/mL. Validated linearity for cell culture lysate (n=5) is 82-98% at 1:2, 84-101% at 1:4, and 90-98% at 1:8 dilution, with recovery of 85-105% (average 95%). A Bradford Reagent is included for total protein normalisation, and the kit is labelled for research-use-only with components stored sealed at 2-8°C without cryopreservation.
