Human PRG-1/LPPR4 ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA kit is designed for the accurate and sensitive detection of PRG-1 (also known as LPPR4, Brain-specific phosphatidic acid phosphatase-like protein 1, or Plasticity-related gene 1 protein) in human samples including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. PRG-1, or Lipid phosphate phosphatase-related protein type 4, is a key protein involved in regulating lipid signaling pathways and plays a critical role in cell migration, adhesion, and proliferation. Abnormal PRG-1 expression has been associated with various diseases, including cancer and neurological disorders, making it an important biomarker for understanding disease mechanisms.

The assay uses a colorimetric detection method in a Sandwich ELISA, Double Antibody format, with a sensitivity of 0.188 ng/mL and a range of 0.313–20 ng/mL. Linearity has been demonstrated in serum (95–101% at 1:2, 87–102% at 1:4, 99–104% at 1:8), EDTA plasma (82–98% at 1:2, 84–100% at 1:4, 82–100% at 1:8), and heparin plasma (84–98% at 1:2, 91–94% at 1:4, 81–99% at 1:8). Recovery ranges are 85–102% (average 94%) for serum, 85–98% (average 93%) for EDTA plasma, and 88–104% (average 98%) for heparin plasma. The kit should be stored at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents to room temperature, loading standards, samples and controls onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labeled detection antibody is then added with a 60-minute incubation, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes before stopping the reaction and measuring absorbance at 450 nm. Sample preparation protocols are provided for serum (centrifuge at 1000 × g for 20 minutes), plasma (centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate.
